Whats the difference between Depo provera and Spironolactone?
Does anyone know the effects that any of these drugs have on the liver?

I'm not too knowledgable on Depo Provera, but it does block testosterone and may also interfere with estrogen receptors as well. Spironolactone is a potasium sparing diuretic which has the side effect of also blocking testosterone.

Anything that is ingested will make two passes through the liver. Once when it is absorbed by the digestive tract, and again when it is excreted. Depo, or intramuscular, will not make this first pass through the liver. Hense, there is less work the liver must perform.
......
Possibly it is not. Delestrogen will peak after it is injected then taper off. Think of it like a roller coaster, there is a great rise at first and as time goes on it drops off. Estradiol tablets will give a consistant dose of estrogens. Again, with the tablets it's making two passes through the liver but it's effects can be increased by taking it sublingualy (under the tongue) and at the same time reducing stress on the liver.
Delestrogen hasn't been proven to be any more effective than tablets, but it is more expensive and does offer the convenience of only having to inject it on a bi-weekly basis.

DP blocks the production of gonadotropin. G causes the body to make both male & female hormones.
