The prostate is approximately .75 inches in from the rectum muscle lining. Obviously this varies between people.

Trying to destroy it through insertion of a single finger is not likely...hard to squeeze it with only one finger. But, constant massaging of it can cause bruising if done hard enough.

Now, if you were to use many fingers and stretch the person open enough you might gain leverage with multiple fingers to be able to span the gland from side to side. The gland is normally approximately the size of a walnut, and can be a bit larger if sexually aroused. You could in theory squeeze the gland once you had that kind of leverage.
